Electrophoresis
current is unavoidable in gel electrophoresis process. Massive heat, which is
generated inside gel matrix, will cause problem to the quality of gel
electrophoresis. Different electrophoresis buffer conducts electrophoresis
current at different level. TAE buffer, for example, conducts higher
electrophoresis current than TBE buffer, due to its higher conductivity (lower
resistance).
High temperature,
elevated by electrophoresis current, reduces sample resolution capacity of gel
electrophoresis. Uneven temperature distribution causes gel smiling.
