How
to prepare standard 50X TAE Buffer & to make your TAE buffer smarter?
TAE buffer has its advantages over TBE buffer. Unlike TBE
buffer, you can prepare TAE buffer at 50x concentrated TAE buffer without
worrying about precipitation.
50X TAE Buffer
To make 2 Liter of TAE buffer, add
484g Trisbase, 200mL of 0.5M EDTA pH 8.0, 114.2mL Glacial Acetic Acid
Add enough MQ H2O to dissolve solids, then bring up final volume to 2000mL
To make 1 Liter of standard 50x TAE buffer, use a 1000ml bottle with 600ml
deionized water and add:
242g Trizma Base, 100mL of 0.5M EDTA pH 8.0
57.2mL Glacial Acetic Acid, then bring final volume to 1000mL.
